چکیده
A full-length cDNA clone encoding rabbit liver arylamine *• N-acetyltransferase (NAT; EC 2.3.1.5) was isolated from a Xgtll [ expression library by concomitant screening with an oligonucleof tide deduced from partial amino acid sequences (1) and with antibodies raised against the purified protein. The nucleotide p sequence of the clone, designated rnat, consists of 1102 nucleotides and contains a single open reading frame of 870 nucleotides A encoding a 290 amino acid protein (Mr 33,655 Da) with 14 of 16 tryptic peptide sequences recently determined from purified * rabbit liver NAT (1). The rnat cDNA shows 61% and 46% sequence homology at the nucleotide and deduced amino acid level, |f respectively, to those of NAT from chicken liver (2).

Molecular and genetic analyses of arylamine N-acetyltransferase polymorphism of rabbit liver.
A cDNA clone encoding the full coding region of polymorphic arylamine N-acetyltransferase was isolated from rabbit liver and expressed in Chinese hamster ovary cells. The expressed enzyme acetylated 2-aminofluorene, procainamide, sulfamethazine, and p-aminobenzoic acid at equivalent rates. N-Acetyltransferase activity was measured in 17 rabbits from an inbred colony which were classified into r...

To elucidate the molecular basis of human N-acetylation polymorphism, cDNA clones encoding human liver N-acetyltransferases (EC 2.3.1.5) were isolated from lambda gt10 cDNA libraries using the 32P-labeled cDNA of rabbit liver N-acetyltransferase recently cloned in this laboratory. Three types of cDNAs (D-14, O-7, and D-24) were isolated and their nucleotide sequences were determined, from which...
متن کاملHuman Liver Arylacetamide Deacetylase MOLECULAR CLONING OF A NOVEL ESTERASE INVOLVED IN THE METABOLIC ACTIVATION OF ARYLAMINE CARCINOGENS WITH HIGH SEQUENCE S I M I M I ’ N TO HORMONE-SENSITJYE LIPASE*
Microsomal arylacetamide deacetylase (DAC) competes against the activity of cytosolic arylamine N-acetyltransferase, which catalyzes one of the initial biotransformation pathways for arylamine and heterocyclic amine carcinogens in many species and tissues. Activity determination and immunoblot analysis of DAC in human target tissues for arylamine carcinogens revealed that in extrahepatic tissu...

The New Zealand White rabbit provides a widely used animal model for the human acetylation polymorphism, which confers marked interindividual variation in the effect and toxicity of numerous drugs, chemicals, and potential carcinogens. The relationship of a recently isolated cDNA clone, designated rnat, to genetically polymorphic arylamine N-acetyltransferase (NAT; acetyl-CoA:arylamine N-acetyl...
متن کاملArylamine N-acetyltransferase from chicken liver. I. Monoclonal antibodies, immunoaffinity purification, and amino acid sequences.
Five monoclonal antibodies against arylamine acetyltransferase (EC 2.3.1.5) from the chicken liver were established by immunizing a mouse with a partially purified enzyme preparation. None of the antibodies cross-reacted with arylamine N-acetyltransferase from the livers of cow, rabbit, and rat, nor with arylalkylamine N-acetyltransferase from the chicken pineal gland, indicating a high specifi...
